Zusammenfassung
SecondHarmonicGenerationMikroskopieinbiomedizinischenAnwendungen
Frequenzverdopplung (SHG) von Licht kann durch verschiedene wichtige Proteinstruktu-
ren in biologischen Proben selbst hervorgerufen werden. Zu diesen zählen vor allem das
extrazelluläreMatrixproteinCollagenunddasMotorproteinMyosininderquergestreiften
Muskulatur.MitHilfevonKurzpulslasernundLaser-ScanningMikroskopiekönnendieSHG
Signaledazugenutztwerden,umdieseProteinstrukturenohnedenZusatzexternerMarker
hochauflösenddarzustellen.
DasHauptzieldieserArbeitisteinebiomolekulareundphysikalischeCharakterisierungdes
SHGSignalsvonMuskelpräparatenundeineDiskussionvonAnwendungsmöglichkeitenan
derHautimKontextvonHautkrebsundSklerodermie.AmMuskelwurdespezielldieOri-
entierungsabhängigkeitdesSHGSignalsvonderPolarisationdesLasersuntersucht.Kom-
plementärdazuwurdentheoretischeModellezurquantitativenBeschreibungundzumVer-
ständnisderbeobachtetenEffekteentwickelt.ErstmalswurdehieraucheinModellderVer-
teilungdeselektrischenFeldesimObjektivfokusindieBerechnungenmiteinbezogen.Das
zentraleErgebnisdieserExperimenteist,dassdieSHGPolarisationsabhängigkeitimMuskel
in den physiologischen Zuständen Relaxed und Rigor unterschiedlich ist. Auf diese Weise
lässtsichalsoderfunktionalemolekulareZustandderMyosinmoleküleausderSHGSignal-
charakteristikablesen.
AnderHautkonntedurchSHGundAutofluoreszenzSignaledieStrukturderoberstenHaut-
schichteninvivomithoherAuflösungdreidimensionalabgebildetwerden.DieSHGMikro-
skopie verspricht eine besondere Eignung als nicht-invasives Werkzeug zum Studium von
Hauterkrankungen.
Abstract
SecondHarmonicGenerationMicroscopyinBiomedicalApplications
Several biologically relevant protein structures are known as sources for second harmonic
generation(SHG)signals.Amongthesearecollagen,anextracellularmatrixprotein,andthe
motorproteinmyosininstriatedmuscle.Incombinationwithshort-pulselasersandlaser-
scanningmicroscopy,SHGsignalscanprovidehigh-resolutionimagingofthesestructures
withouttheneedforextrinsiclabels.
Themainobjectiveofthisstudyistocharacterizethephysicalandbiomolecularproperties
of SHG from mammalian skeletal muscle and further to discuss applications of SHG mi-
croscopytotheskininthecontextofskincancerandscleroderma.Inmuscle,aspecialfocus
layontheSHGorientationdependencyontheaxisoflaserpolarization.Complementaryto
theexperimentalwork,severaltheoreticalmodelsweredevelopedforaprecisequantitative
descriptionandadeeperunderstandingoftheobservedeffects.Forthefirsttime,theentire
electricfielddistributioninthefocusoftheobjectivewasconsideredhereforthemodeling
ofthepolarizationdependency.Thecentralresultoftheconductedexperimentsisthatthe
SHGpolarizationdependencyisdifferentforthetwophysiologicalstatesrelaxed andrigor
inmuscle,whichmeansthatthefunctionalmolecularstateofthemyosinmoleculesisex-
pressedintheSHGsignalcharacteristics.
Inmurineskin,SHGandautofluorescencesignalsshowedthethree-dimensionalstructure
of the upper skin layers in vivo with high resolution. SHG microscopy proposes excellent
suitabilityasanon-invasivetoolforstudyingcutaneaousconditions.
