Preface
The enzymology of phospholipid biosynthesis was covered as a small
section in Volume 17 of Methods in Enzymology published in 1981. In the
past decade, there have been many rapid advances in the purification and
cloning of the enzymes involved in phospholipid biosynthesis. In 1981,
only a few of the enzymes described had been purified and none cloned or
sequenced.
This volume reports on the purification and cloning of many of the
phospholipid biosynthetic enzymes from bacteria, yeast, and animal tis-
sues. In addition, new strategies have been developed for the selection of
mutants defective in phospholipid biosynthesis. Moreover, there has been
significant progress in closely related areas of research that impinge on
phospholipid metabolism: platelet activating factor, sphingolipid biosyn-
thesis, phospholipid transfer proteins, and lipid A biosynthesis in Escheri-
chia coli. Thus, it is timely and important to collate in one volume the new
methods and procedures for studies of the enzymes of phospholipid bio-
synthesis.
This book will be a useful reference source for scientists in the field
and will be a catalyst for further advancements on the structure and
function of the enzymes of phospholipid biosynthesis. It should also be
highly useful for studies on the regulation of phospholipid metabolism and
the elucidation of the function of phospholipids.
This book is dedicated to Professor Eugene P. Kennedy, Hamilton
Kuhn Professor Emeritus at Harvard Medical School, who discovered the
central de novo pathways for the biosynthesis of phospholipids involving
cytidine nucleotides and who has continued to make seminal contribu-
tions to advancing the understanding of phospholipid and membrane bio-
chemistry and enzymology.
